The instrument can be used to measure the light scattering and fluorescence parameters of large cell populations on a cell-by-cell basis fast, precisely and reproducibly. The light source is a 488-nm Ar ion laser. Measureable parameters: forward and side scattering, fluorescence intensities of 3 spectrally different dyes. With these parameters infromation on the following cellular properties can be gained: expression of intracellular and cell surface proteins (using immunefluorescent labeling); cell viability (calcein-AM, FDA and/or propidium-iodide); intracellular pH (BCECF-AM); membrane potential (oxonol); DNA-content, which can be used to characterize the cells within different phases of the cell cycle; intracellular accumulation of fluorescent substrates of such as those of ABC transporters (e.g. daunorubicin, doxorubicin, calcein, rhodamine 123).
