About the facility

The RI serves the Life Science Center of the UD with confocal microscopy measurements required for research on morphology, cell biology and physiology research. Samples from clinical research are measured as well. As one of the 2 FCS instruments in the country, it is able to measure diffusion constant and concentration in one measurement with high sensitivity. This modality makes possible to study stable molecular interactions in intracellular subcompartmental volumes.

Zeiss LSM 510 - Confocor 2 confocal laser scanning microscope equipped with two-channel fluorescence correlation/crosscorrelation spectroscope. Equipped with four Laser lines: 351, 364, 458, 477, 488, 514 543,633 nm. 4 channel simultaneous detection, one is capable of spectral imaging with META detector (377-720 nm, max. 8 channels, BP: 10,7-42,8 nm) Transmission channel for taking pictures similar to conventional brightfield microscopes. Capable of taking "optical slices" showing thin layers of the sample and reconstruct a 3D image from these slices. Mosaic images of large area samples (histological samples) can be taken. Capability of measuring ion concentrations on a millisecond scale using line scanning and fluorescent indicators. With the ConfoCor2 fluorescence correlation detector it is possible to measure fluorescence fluctuation, auto and crosscorrelation in an approx. 0,25 femtoliter volume. The diffusion constant of the fluorescent molecules, molecular interactions and association states can be calculated.
