About the facility

The Olympus FV-1000 confocal microscope can make 3D fluorescence images of live or fixed cells and tissues with a resolution of 200 nm. The setup has 3 fluorescence detectors (2 with spectral resolution) + DIC. Lasers: Ar ion (457, 488, 514 nm), green HeNe (543 nm), red HeNe (633 nm). Planned extension: 405 nm laser. Fluorescence correlation spectroscope (FCS) attached to the confocal microscope Auto- or crosscorrelation measurements can be carried out in two detection channels (APD detectors) at selected points of the confocal image or in solution. Measurable parameters: local diffusion (mobility), local concentration (sensitivity: 100 pM, <1 molecule in the confocal volume), aggregation state, co-mobility, stable homo/hetero-association. Development of single molecule FRET technique is in progress. The instrument operates under the aegis of the Molecular Cell Analytic Core Facility.
